Mechanism of Liver Damage Prevention and Interleukin-6 Decrease by Red Onion Extract: A Partial Least Squares Analysis in Diabetic Rats

Abstract

Worldwide, in 2025, it is estimated that 350 million people will suffer from diabetes mellitus. The presence of dysfunction and changes in hepatic cell mass indicates histological changes in the liver and triggers the IL-6 inflammatory process. The study aims to analyze the mechanism by which red onion extract prevents liver damage and reduces the level of IL-6. In this study, 24 white rats were divided into six groups. Five of the six groups were diagnosed as diabetic and given intraperitoneal alloxan injection. The study groups consisted of six groups: control group without diabetes (A); alloxan-induced group without therapy (B); alloxan-induced group treated with synthetic drug metformin dose of 45 mg/kg (C); alloxan-induced group treated with red onion extract dose 1 (D); alloxan-induced group treated with dose 2 (E); alloxan-induced group treated with dose 3 (F). The fixed hepatic organs were then examined using HE staining and IHC. Statistical analysis of the PLS test. The results showed the highest average necrosis in the group not given red onion extract, at 3,550. Giving red onion extract lowers the mean IL-6 by 2.550 compared to the group that did not receive it. The mechanism of providing red onion extract directly reduces necrosis, and the IL-6 t-value is greater than 1.96. This study concludes that administering red onion extract can prevent liver damage and reduce IL-6 levels. This study can inform the public that red onions may be developed as an alternative treatment for humans with DM. © 2026 LETTERS IN BIOMATHEMATICS. All rights reserved.
